The aim of WP7 is to develop high-throughput technologies for systematic quantitative protein profiling of tissues and biofluid samples from MolPAGE WP1, using the antibodies generated in WP6. These include tissue microarrays, first generation protein arrays, antibody arrays, serum arrays, microfabricated CD devices, and mass spectrometry. The ultimate goal of WP7 is to refine and develop these tools application on an epidemiological scale in the future study of large population cohorts.
The volume of samples available for epidemiological studies can be limited; especially those sourced from longitudinal population cohorts. For this reason one objective for WP7 is to improve the sensitivity of ligand detection when using a small volume of biofluid for biomarker detection within a micro-fabricated CD system for microfluidics
Antibodies generated towards MolPAGE candidate genes (see WP6) will be used to determine the expression of the protein and it’s sub-cellular localization in a panel of 48 normal human tissue sections embedded in a tissue microarray. Images from these immunohistochemistry experiments will be made publicly available via the Human Protein Atlas website; www.proteinatlas.org. As part of the MolSURG study WP1 have procured paraffin embedded metabolically interesting tissues from subjects with and without T2D. WP7 will generate a ‘metabolic tissue microarray’ from these samples for analysis with the MolPAGE antibodies.
